Monoclonal Antibody Purification with Reduced Turbidity by using High-salt Elution Buffer During Protein a Chromatography

During purification of monoclonal antibodies, viral inactivation/removal step is a mandatory to comply with the regulatory guidelines which commonly performs at low pH about 3.5 and further neutralizes it 7.0. Neutralization antibodies results formation high turbidity troublesome for processing such as filtration other chromatography processes eventually leads filter clogging increases column back pressure. In this work, we propose new optimized process affinity (Protein A) by using salt elution buffer reduces more than 80%. A systematic study different concentration salts were analysed reveals that greater 100mM in Protein during neutralization when compared buffer. We also found higher reduction 3.0. These findings provide both cost time effective large scale production eliminates use multiple filters throughout process.
